Abstract

Investigations were carried out in phytopathological and in vitro laboratories of Tobacco Institute-Prilep. A fragment of pure culture of the fungus, suspension prepared from fungus mycelium and filtrate as a selective agent obtained from the fungus grown on fluid nutritive medium were used as inoculum. Fluid medium was filtrated through filter paper and through bacteriological filter of 0,22µ porosity
When tobacco leaves were inoculated with fungus suspension and selective agent applied as a drop on leaf surface, necrosis development was more rapid on the first day, in the variant with pure filtrate, while on the 15th day it was much higher in the suspension prepared from fungus mycelium. The highest development of necrosis in all days of observation was recorded in inoculation with a fragment of pure culture.
By immersing the leaf base in the inoculum, rapid necrosis development in all days of observation was recorded in the selective agent, while in the inoculum of mycelial suspension, this development was slower.
Symptoms on leaf surface caused by toxins of the pathogen did not differ from those caused by the pathogen itself.
The aim of investigation was to check the infectiveness of toxins released by the pathogen and differences in necrosis development depending on the three inoculi applied.
